I use the Web Developer toolbar for Firefox. It has heaps of very useful features to help optimize a new site.

That 2nd link's cool--didn't know about it. Thanks.

I use the Web Developer toolbar, as well. Just validate local XHTML and CSS.
Sometimes I'll go through checking my accessibility, but usually it's pretty minute stuff that it finds, if anything.

Possibly a little extreme:D but it is a very valuable tool (or set of tools) and not just for validation. Being able to change the css on the fly is great

Just tried sitereportcar. Bookmarket straight away, very useful set of reports in one place.
