Is there a simple way to set up my own Wordpress distribution? I'd like to package all my favorite plugins, a couple good themes and upon initial setup everything's enabled and ready to go.

Yes. You take all the files and you put them in their respective folders. Then after installation, you click select all, and activate all. Easy. FFS!
you can hack the setup files, you could copy a db of a fresh install and change specifics for new installations, etc. so yes you can.
